About Glutathione

Glutathione (GSH) is composed of glutamate, cysteine, and glycine, forming one of the body’s primary intracellular antioxidants. Research frequently examines its role in redox balance, detoxification, immune modulation, and mitochondrial protection. GSH participates directly in neutralizing reactive oxygen species and maintaining the cellular redox state through enzymatic cycling with its oxidized form, GSSG. It is a foundational molecule in oxidative stress and cellular defense studies.

FAQs

What is glutathione?
Glutathione is a tripeptide composed of three amino acids: glutamate, cysteine, and glycine. It is the most abundant intracellular antioxidant in mammalian cells and is synthesized endogenously by the enzyme glutamate-cysteine ligase followed by glutathione synthetase. Glutathione exists in reduced (GSH) and oxidized (GSSG) forms, and the ratio of GSH to GSSG is used as a marker of cellular oxidative stress.
Glutathione neutralizes reactive oxygen species through enzymatic and non-enzymatic pathways. As a substrate for glutathione peroxidase, it reduces hydrogen peroxide and lipid hydroperoxides to water and corresponding alcohols. Glutathione also regenerates other antioxidants, including vitamins C and E, back to their active reduced forms. The glutathione S-transferase enzyme family uses glutathione as a cofactor to conjugate and detoxify electrophilic compounds and xenobiotics.
Glutathione is studied in the context of redox homeostasis, phase II detoxification, protein thiol protection, and cell proliferation regulation. Depletion of intracellular glutathione is used experimentally to induce oxidative stress and apoptosis in cell culture models. Researchers measure GSH and GSSG levels using assays such as DTNB-based colorimetric methods, HPLC, and mass spectrometry to assess cellular redox status under various experimental conditions.
Published research reports that intracellular glutathione levels decrease with age in multiple tissues. This decline is associated with reduced activity of the rate-limiting enzyme glutamate-cysteine ligase and increased oxidative burden. Studies in aged animal models show that glutathione depletion correlates with increased lipid peroxidation, protein carbonylation, and DNA oxidation markers, which are hallmarks of age-related cellular damage.
